Analysis of Molecular Variance(AMOVA) is a method for studying molecular variation within a species. Electrophoresis, one of the most widely used methods for studying the structure of DNA, produces data of the form of 0s and 1s where 1 denotes the presence of a band and zero its absence, e. g. Sample Band A 0101... B 1000... C 0110... AMOVA works on such data to create a distance matrix between samples in order to measure the genetic structure of a population. One of the recommendations of the panel that has attracted the attention of attorneys is that fingerprinting tests showing a phenomenon called band shifting be declared "inconclusive" until laboratories have performed adequate studies on the effect. Band shifting can make two forensic samples from different biological sources appear the same; private companies that perform DNA fingerprinting and the Federal Bureau of investigation have already made attempts to correct for it. Fluorescence-based amplified fragment length polymorphism(fbAFLP) is a novel assay based on the fluorescent analysis of an amplified subset of restriction fragments. The fbAFLP assay involves the selective PCR [polymerase chain reaction] amplification of restriction fragments from a total digest of genomic DNA. The ligation of adapters with primer-specific sites coupled with primers containing selective nucleotides allowed the full potential of PCR to be realized while maintaining the advantages of restriction endonuclease analysis.
