A physical mapping approach that uses fluorescein tags to detect hybridization of probes with metaphase chromosomes and with the less-condensed somatic interphase chromatin.

Until recently, even the best chromosomal maps could be used to locate a DNA [deoxyribonucleic acid] fragment only to a region of about 10 Mb, the size of a typical band seen on a chromosome. Improvements in fluorescence in situ hybridization(FISH) methods allow orientation of DNA sequences that lie as close as 2 to 5 Mb.

Fluorescent in situ hybridization on interphase nuclei prepared from normal male fibroblast cultures gave a distance between D9S12 and D9S180 of about one Mb.

Analysis of Molecular Variance(AMOVA) is a method for studying molecular variation within a species. Electrophoresis, one of the most widely used methods for studying the structure of DNA, produces data of the form of 0s and 1s where 1 denotes the presence of a band and zero its absence, e. g. Sample Band A 0101... B 1000... C 0110... AMOVA works on such data to create a distance matrix between samples in order to measure the genetic structure of a population. In statistical terms, AMOVA is a testing procedure based on permutational analysis and involves few assumptions.

Les chromosomes métaphasiques humains présentent divers types de bandes selon le traitement auquel ils sont soumis (colorants, température, enzymes protéolytiques ou DNases). Les bandes G(iemsa) positives représentent environ 50 % des bandes chromosomiques, l'autre moitié étant constituée de bandes G(iemsa) négatives. Ces dernières ne sont que les bandes R(everse) obtenues par dénaturation thermique.

One of the recommendations of the panel that has attracted the attention of attorneys is that fingerprinting tests showing a phenomenon called band shifting be declared "inconclusive" until laboratories have performed adequate studies on the effect. Band shifting can make two forensic samples from different biological sources appear the same; private companies that perform DNA fingerprinting and the Federal Bureau of investigation have already made attempts to correct for it. But the McKusick panel was not convinced that such attempts protect those accused : a suspect could be wrongly convicted if the "correction" was itself in error.

Fluorescence-based amplified fragment length polymorphism(fbAFLP) is a novel assay based on the fluorescent analysis of an amplified subset of restriction fragments. The fbAFLP assay involves the selective PCR [polymerase chain reaction] amplification of restriction fragments from a total digest of genomic DNA. The ligation of adapters with primer-specific sites coupled with primers containing selective nucleotides allowed the full potential of PCR to be realized while maintaining the advantages of restriction endonuclease analysis. Fluorescence-based fragment analysis with polyacrylamide gel electrophoresis provides the accurate band sizing required for homology assessment.

FISH analysis using chromosome 8-specific centromeric alphoid probe and C-banding further demonstrated that the minichromosome is derived from chromosome 8 centromere... Southern blot analysis using Ecogpt gene as a probe after pulsed-field gel electrophoresis of high molecular weight DNA in agarose plugs did not display any hybridization. However, hybridization of blots containing gamma-irradiated DNA or Not1 digested DNA in agarose plugs with the Ecogpt gene probe showed a 1. 6 mb band suggesting circular nature of the minichromosome.
