A recombinant form of Human Interferon Beta made by Triton Biosciences designed to treat HIV infection and Kaposi’s Sarcoma. It is classified as: antineoplastic, antiviral, immunomodulator, antiretroviral.

BETASERON : Berlex Canada, interferon beta-1b is an immunomodulator used in ambulatory patients with relapsing-remitting multiple sclerosis. Betaseron is a highly purified, sterile, lyophilized protein product produced by recombinant DNA techniques and formulated for use by injection. It is manufactured by bacterial fermentation of a strain of E. coli that bears a genetically engineered plasmid containing the gene for human interferon betaser17. The native gene was obtained from human fibroblasts and altered in a way that substitutes serine for the cysteine residue found at position 17.

The pCOR plasmids are narrow-host range plasmid vectors for nonviral gene therapy. These plasmids contain a conditional origin of replication and must be propagated in a specifically engineered E. coli host strain, greatly reducing the potential for propagation in the environment or in treated patients.

Purification of mutant enzymes is a prime requirement of biophysical and biochemical studies. Our investigations on the essential E. Coli enzyme glutaminyl-tRNA sybthetase demand mutant enzymes free of any wild-type protein contamination. However, as it is not possible to express non-complementing mutant enzymes in an E. Coli gInS-deletion strain, we developed a novel strategy to address these problems. Instead of following the common tactic of epitope-tagging the mutant protein of interest on an extrachromosomal genetic element, we fused a reporter epitope to the 5’ end of the chromosomal gInS-gene copy : this is referred as reverse epitope-tagging.

A good example of the [DNA recombinant] technique comes from work carried out by ICI where a gene for an energy conserving nitrogen pathway taken from E. coli was transferred into their SCP(single-cell protein) organism Methylophilus methylotrophus strain AS1 where it substituted for the similar but energy consuming pathway possessed by the strain.
