ELISPOT(Enzyme-linked ImmunoSPOT) technique was originally used to enumerate antibody secreting B cells. In the current technique, cells are deposited onto a membrane coated with one antibody specific for a protein followed by an appropriate incubation period. Subsequently, the protein of interest is detected in the environment immediately surrounding the cell secreting it, with another antibody specific for a different epitope of the protein.

Purification of mutant enzymes is a prime requirement of biophysical and biochemical studies. Our investigations on the essential E. Coli enzyme glutaminyl-tRNA sybthetase demand mutant enzymes free of any wild-type protein contamination. However, as it is not possible to express non-complementing mutant enzymes in an E. Coli gInS-deletion strain, we developed a novel strategy to address these problems. Instead of following the common tactic of epitope-tagging the mutant protein of interest on an extrachromosomal genetic element, we fused a reporter epitope to the 5’ end of the chromosomal gInS-gene copy : this is referred as reverse epitope-tagging.

A recently developed technology called epitope scanning permits the rapid and accurate delineation of continuous stretches of amino acids in a protein which constitute the sequential epitopes recognised by an antiserum raised to that protein. In the present report, we describe the use of this technique to identify the epitopes in the recombinant bovine growth-hormone(rbGH) molecule recognised by three polyclonal guinea-pig antisera and two polyclonal rabbit antisera. The results obtained show that, for guinea-pig antisera, 3 or 4, very-well-defined major continuous epitopes are present.
