Confocal microscopy. Unlike "normal" microscopy, only a spot of a minimum size in the specimen is illuminated in confocal microscopy. To enable an image to be formed from this, the specimen must be scanned. The image of the spot is directed through a pinhole stop in an intermediate image plane. As a result, only light from the focal plane can reach the detector(a photomultiplier). All other(out-of-focus) planes are blocked out. This results in an "optical section". The images are stored electronically and displayed on a monitor. A series of optical sections can be recorded by moving a motor a slight distance along the z-axis each time an image has been recorded, after which the next image is then also recorded. Such a z-series permits the electronic reconstruction of the three-dimensional structure using suitable computer programs. STED [stimulated emission depletion] microscopy was the first high-resolution microscopy concept applied more widely for the study of biological questions. As performed in classical confocal laser scanning microscopy, a conventional laser beam is used to stimulate the fluorescent molecules in what is a normal excited spot(of several hundred nanometers in diameter). A second toroidal(doughnut shaped) beam is used to quench the fluorophores by stimulated emission. By overlapping the two beams, fluorescence is allowed only from the center of the spot, which thus generates a much smaller exciting focal spot... Thus, in practical terms, a smaller effective PSF [point spread function] is generated that causes less blurring and, therefore, results in higher resolution. By applying compression to only a specific area of the breast, the effective pressure can be increased on that spot. This results in better tissue separation and allows better visualization of the small area in question. Laser Scanning Confocal Microscopy(LSCM, also referred to as CSLM, Confocal Scanning Laser Microscopy) is now established as a valuable tool for obtaining high resolution images and 3-D reconstructions of a variety of biological specimens. In LSCM, a laser light beam is expanded to make optimal use of the optics in the objective. Through a x-y deflection mechanism this beam is turned into a scanning beam, focussed to a small spot by an objective lens onto a fluorescent specimen. The mixture of reflected light and emitted fluorescent light is captured by the same objective and(after conversion into a static beam by the x-y scanner device) is focused onto a photodetector(photomultiplier) via a dichroic mirror(beam splitter). The reflected light is deviated by the dichroic mirror while the emitted fluorescent light passes through in the direction of the photomultiplier. A confocal aperture(pinhole) is placed in front of the photodetector, such that the fluorescent light(not the reflected light!) from points on the specimen that are not within the focal plane(the so called out-of-focus light) where the laser beam was focussed will be largly obstructed by the pinhole.
