Short tandem repeat polymorphisms, also referred to as variable number of tandem repeats(VNTRs), are very important to scientists, because they can be used to identify individuals genetically. Marker systems are used to detect loci that respond to restriction enzyme cleavage by producing a fragment that can have many different lengths. This polymorphism is due to variation in the number of tandem repeats. VNTRs can be identified in various ways. Probes can be developed which will hybridize to complementary sequences after RFLP has been performed on a sample of genomic DNA(Southern blots). A second method involves the use of PCR. If the sequence surrounding the VNTR is known, PCR primers(oligonucleotides) can be designed to amplify the repeat region(s). My research interests may broadly be defined as encompassing both "pattern" and "process" aspects of plant evolutions. Chromosome walking, one strategy for filling in gaps, involves hybridizing a primer known sequence to a clone from an unordered genomic library and synthezing a short complementary strand(called walking along a chromosome). The complementary strand is then sequenced and its end used as the next primer for further walking; in this way the adjacent, previously unknown region is identified and sequenced. Two strategies have been used to clone specific sequences of eukaryotic, genomic DNA in bacteriophage lambda vectors. The first involved cloning of DNA preparations that had been highly enriched for the sequences of interest. Total genomic DNA was digested to completion with a restriction endonuclease and the resulting fragments were separated by preparative gel electrophoresis, reverse-phase column chromatography, or both. The fractions containing the DNA sequences of interest were identified by hybridization to appropriate probes and cloned in bacteriophage lambda vectors. Because these fractionation procedures may yield an overall enrichment of as much as 100-fold to 300-fold, the number of recombinant bacteriophages that had to be generated and screened for the DNA sequences of interest could be significantly reduced. Subsequently, the development of efficient in vitro packaging systems and in situ hybridization techniques eliminated the need to enrich the starting DNA for the sequences of interest. 1, record 67, English, - reverse%20phase%20column%20chromatography
