DNA [deoxyribonucleic acid] segments defined by helitron termini contained multiple gene-derived fragments and had a structure typical of nonautonomous helitron-like transposons. Genomic DNA is maintained with high fidelity to guarantee the integrity of genetic information. Damaged DNA must be properly repaired by sophisticated DNA repair systems to sustain cell viability and genomic stability. Homologous recombination is the major pathway for repairing DNA double strand breaks and for re-initiating stalled replication forks in E. coli and eukaryotes. In E. coli, there are several important proteins involved in homologous recombination : RecA protein is essential for pairing homologous DNA, and for promoting the strand exchange process. 2, record 9, English, - recA%20protein

1, record 9, French, - prot%C3%A9ine%20recA

Short tandem repeat polymorphisms, also referred to as variable number of tandem repeats(VNTRs), are very important to scientists, because they can be used to identify individuals genetically. Marker systems are used to detect loci that respond to restriction enzyme cleavage by producing a fragment that can have many different lengths. This polymorphism is due to variation in the number of tandem repeats. VNTRs can be identified in various ways. Probes can be developed which will hybridize to complementary sequences after RFLP has been performed on a sample of genomic DNA(Southern blots). A second method involves the use of PCR. If the sequence surrounding the VNTR is known, PCR primers(oligonucleotides) can be designed to amplify the repeat region(s). The information obtained from these experiments is useful in the fields of forensics, population characterization, human gene mapping, identification of genes involved in disease, relationship testing, and identifying genes of interest such as those that encode proteins containing zinc finge. 3, record 13, English, - variable%20number%20of%20tandem%20repeats

2, record 13, French, - nombre%20variable%20de%20r%C3%A9p%C3%A9titions%20en%20tandem

The most important aspect of our cloning vectors is that they are based on the E. coli F-factor replicon. It allows for strict copy number control of the clones so that they are stably maintained at 1-2 copies per cell. The stability of the cloned DNA during propagation in E. coli host is substantially higher in lower-copy number vectors than in multi-copy counterparts.... The stabilizing effect of BAC and cosmid vectors is notable especially for certain genomic DNA that are normally unstable in high copy number vectors. 1, record 16, English, - bacterial%20artificial%20chromosome

Fluorescence-based amplified fragment length polymorphism(fbAFLP) is a novel assay based on the fluorescent analysis of an amplified subset of restriction fragments. The fbAFLP assay involves the selective PCR [polymerase chain reaction] amplification of restriction fragments from a total digest of genomic DNA. The ligation of adapters with primer-specific sites coupled with primers containing selective nucleotides allowed the full potential of PCR to be realized while maintaining the advantages of restriction endonuclease analysis. Fluorescence-based fragment analysis with polyacrylamide gel electrophoresis provides the accurate band sizing required for homology assessment. 5, record 17, English, - fluorescent%20amplified%20fragment%20length%20polymorphism

1, record 20, French, - PCR%20%C3%A9lectronique

Breast cancer progression is determined by a complex pattern of multiple genetic aberrations the association of which with patient prognosis is unknown. In this study, we have undertaken a genome-wide screening to detect genetic changes associated with clinical outcome in node-negative breast cancer. Comparative genomic hybridization was used to screen for DNA sequence gains and losses across all human chromosomes in 23 tumors... 1, record 23, English, - genomic%20hybridization

Two strategies have been used to clone specific sequences of eukaryotic, genomic DNA in bacteriophage lambda vectors. The first involved cloning of DNA preparations that had been highly enriched for the sequences of interest. Total genomic DNA was digested to completion with a restriction endonuclease and the resulting fragments were separated by preparative gel electrophoresis, reverse-phase column chromatography, or both. The fractions containing the DNA sequences of interest were identified by hybridization to appropriate probes and cloned in bacteriophage lambda vectors. Because these fractionation procedures may yield an overall enrichment of as much as 100-fold to 300-fold, the number of recombinant bacteriophages that had to be generated and screened for the DNA sequences of interest could be significantly reduced. Subsequently, the development of efficient in vitro packaging systems and in situ hybridization techniques eliminated the need to enrich the starting DNA for the sequences of interest. 1, record 26, English, - reverse%20phase%20column%20chromatography
