In another application of gene therapy we have used an adenoviral vector that carries the gene for herpes simplex virus thymidine kinase(HSV-tk). Cells expressing HSV-tk will convert the anti-viral drug ganciclovir(GCV) to a form that is toxic to rapidly dividing cells. Using this adenoviral vector we have repeatedly shown robust tumoricidal effects in a variety of animal models of CNS tumor and will soon take this treatment to phase I clinical trials. An ingenious experiment offers hope for a new kind of surgery for cancer. Kenneth Culver(National Cancer Institute-USA spliced a snippet of DNA from a herpes virus into one of these retroviruses and injected the combination into rats suffering from brain cancer. Since cancer cells are about the only cells that are dividing in a cancer-infected brain, the viruses were supposed to invade those cells and multiply. After five days, the rats were treated with an antiherpes drug. The hope was that the toxin would kill herpes-infected cancer cells and leave the rest of the brain alone. In 11 of the 14 rats, the tumors disappeared completely. The results were so promising that an NIH watchdog committee has already approved a similar test on humans. The risks are high. The researchers will, in effect, be putting mouse genes directly into human brains. But the payoff could be great. Feline viral rhinotracheitis: an acute upper respiratory infection of cats, caused by feline herpesvirus. Sneezing, coughing, ocular and nasal discharges, and often chemosis, keratitis and corneal ulceration are the main signs. Occasionally there is fatal pneumonia or generalized infection in young kittens. The colloquial name for an illness of puppies, leading usually to their death within a few days of birth. Symptoms include : progressive weakness which soon makes suckling impossible; a falling body temperature; and "paddling" movements. Infected puppies may be killed by their dams.
