When one wishes to perform site specific mutagenesis on a gene and re-express the mutant genes into altered proteins, several considerations should be taken initially to increase the efficiency of generating mutants. The first step is to construct the gene by chemical synthesis as it allows for the alteration of nucleotide sequence to accommodate changes in the overall design. In the case where host restriction... endonuclease recognition sites of the natural cDNA sequences are not suitable for performing molecular cloning, then it is possible to create additional useful restriction endonuclease recognition sites or alter the pre-existing sites. Because of the degeneracy of the genetic code, a particular amino acid can be specified by more than one, sometimes up to 6 different codons.... A computer program has been devised... which would search upon all these possible combinations of codons, while keeping the original amino acid sequence. 3, record 7, English, - chemical%20synthesis

A large city of Alberta, located on the Bow River, in the southern part of the province, at the meeting of the Canadian prairies and the Rocky Montains foothills. The city started as a trading post(fur trade, bison hunting and illicit-whisky trading) with the construction of a fort named Fort Calgary by the North-West Mounted Police at the confluence of the Bow and Elbow rivers in 1876.(The word Calgary, of Gaelic origin, means "bay farm. ") "Fort Calgary" is linked to Canada by the railway in 1883; the Canadian Pacific Railway(CPR) Company laid out its town site where the city now lies. Incorporated under the name "Calgary" in 1884, the town received the city status in 1893. Financial centre of Western Canada and headquarters of Canada's oil and natural gas industries, Calgary, host of the 1988 Winter Olympic Games, is the fastest growing Canadian city in the West. 2, record 10, English, - Calgary

2, record 10, French, - Calgary

The Common Departmental Financial System(CDFS) is a comprehensive on-line departmental financial management and reporting system designed to simplify and streamline financial administration. CDFS was developed to respond to the changing needs for financial management information in the federal government. It complements recommendations outlined in Public Service 2000 which called for departmental line managers to assume a greater responsibility for financial management of the areas under their control. Offered by Public Works and Government Services Canada(PWGSC), CDFS is an optional service. It replaces the FINCON and FINCAP systems. CDFS is a distributed system. Under the current configuration, the PWGSC Mainframe acts as a host while the presentation layer resides at the client site on a Local Area Network(LAN) or workstation. 2, record 14, English, - Common%20Departmental%20Financial%20System

2, record 14, French, - Syst%C3%A8me%20financier%20minist%C3%A9riel%20commun

Experiments have demonstrated that the level of gene expression is influenced principally by the number of gene copies and the efficiency of their transcription and translation. Transcription of a cloned sequence requires the presence of a promotor recognised by the host RNA polymerase and efficient translation requires that the mRNA bears an appropriate ribosome binding site and a correct translational reading frame. 1, record 23, English, - translational%20reading%20frame
