In the case of allogeneic transplants, identifying a potential donor who can give their stem cells to another person involves analyzing the human leukocyte antigens(HLA), which are proteins found on the surface of the blood and in tissue cells. The HLA must be as closely matched as possible so that the transplant recipient's body can "accept" the new stem cells into their bone marrow. In 1955, Barnes and Loutit first described graft versus host disease(GVHD) in mice. Simonsen introduced the term graft-versus-host reaction in 1958 to describe the direction of the immunological damage caused by introduction of immunologically competent cells into an immunocompromised host. In 1966, Billingham proposed 3 conditions required for the development of GVHD, as follows :(1) the graft must contain immunologically competent cells,(2) the host must possess important transplant alloantigens that are lacking in the donor graft so that the host appears foreign to the graft, and(3) the host itself must be incapable of mounting an effective immunological reaction against the graft. Blood irradiator BI-2000. Applications : Irradiation of blood & blood products by gamma rays is a proven and safe method to inhibit T-Lymphocyte proliferation and eliminate the risk of post transfusion graft versus host disease(T-GVHD). Gamma irradiation of cellular blood components is the best current technology to reduce the risk of T-GVHD to the recipients as confirmed by research and therefore being widely practiced world over in hospitals and blood banks as a life saving approach to this problem. Blood is usually irradiated in standard blood bags in dedicated blood irradiators using cobalt-60 or caesium-137 radioactive source.
