IgG antibodies are found in all body fluids. They are the smallest but most common antibody(75% to 80%) of all the antibodies in the body. IgG antibodies are very important in fighting bacterial and viral infections. IgG antibodies are the only type of antibody that can cross the placenta in a pregnant woman to help protect her baby(fetus).

The first antibodies produced in a humoral immune response are IgM, but activated B cells subsequently undergo isotype switching or class switching to secrete antibodies of different isotypes :IgG [inmunoglobulin G], IgA, and IgE. Isotype switching does not affect antibody specificity significantly, but alters the effector functions that an antibody can engage. Isotype switching occurs by a site-specific recombination involving the deletion of the intervening DNA.

The Bartels PRIMA SystemTM Cytomegalovirus IgM Enzyme Immunoassay is a rapid, qualitative method for the detection of IgM Antibody to cytomegalovirus in human serum. Patient serum is diluted with specimen diluent that contains an absorbent to remove IgG. The absorbent prevents IgG/rheumatoid factor complexes from forming and prevents interfering competition from IgG during the test procedure. The treated serum is incubated with purified CMV antigens bound to EIA plate wells. If antibodies to CMV are present, they bind to the antigen and do not rinse off. Subsequently, when enzyme labeled anti-human IgM is added to the reaction site, it binds to the immobilized antibodies. After washing and the addition of a chromogenic substrate and stop solution, specimens containing antibodies to CMV produce a color endpoint reaction which is measured and compared to negative and positive controls, and may be measured spectrophotometrically.

A method used to detect and enumerate antibody producing cells. Cells obtained from, e. g. the spleen of an animal that has been immunized against sheep erythrocytes, are mixed into molten Agarose containing sheep erythrocytes. The agarose is poured into a Petri dish, allowed to set and then incubated at 37 ° C. Later, complement is poured into the dish, and any cells that have secreted antibody are revealed by the appearance of a zone of haemolysis round each of them. The agarose layer can be dried and stained so that the cells may be identified. The technique described reveals IgM secreting cells forming direct plaques. By the addition of an anti-IgG serum before the complement, indirect plaques due to IgG secreting cells can be revealed. Cells producing antibodies to antigens other than erythrocytes can be detected by coating the rythrocytes with the antigen as in passive haemolysis.
