Confocal microscopy. Unlike "normal" microscopy, only a spot of a minimum size in the specimen is illuminated in confocal microscopy. To enable an image to be formed from this, the specimen must be scanned. The image of the spot is directed through a pinhole stop in an intermediate image plane. As a result, only light from the focal plane can reach the detector(a photomultiplier). All other(out-of-focus) planes are blocked out. This results in an "optical section". The images are stored electronically and displayed on a monitor. A series of optical sections can be recorded by moving a motor a slight distance along the z-axis each time an image has been recorded, after which the next image is then also recorded. Such a z-series permits the electronic reconstruction of the three-dimensional structure using suitable computer programs. The procedure, which is restricted to incident-light techniques, has completely revolutionized fluorescence microscopy in biology in particular.

Un des aspects les plus intéressants de la microscopie confocale, outre son caractère non destructif, est sa rapidité de mise en œuvre; elle comble un fossé important entre la microscopie optique conventionnelle et la microscopie électronique. La localisation de molécules spécifiques, dans le contexte cellulaire, apporte d'importantes contributions à l'étude fonctionnelle des systèmes biologiques à différents niveaux d'organisation : subcellulaire, cellulaire, tissulaire ou organisme entier. L'utilisation de traceurs fluorescents très spécifiques permet de localiser in situ avec une résolution spatiale de l'ordre de 400 - 500 nanomètres, des composants macro-moléculaires tels que acides nucléiques et protéines, mais aussi de petites molécules telles que des peptides, lipides et même des ions.

La microscopie confocale permet la localisation in situ d'une sonde fluorescente dans des cellules d'un spécimen d'une épaisseur de 10 mm à 50 mm selon certains auteurs (5 mm à 30 mm plus raisonnablement), sans sectionnement mécanique préalable. Son concurrent est le laser krypton (longueur d'onde) qui permet une excitation des fluorochromes situés dans le Vert (Texas-Red, Cyanin Blue).

Single Particle Analysis is a group of related computerized image processing techniques used to analyze images from transmission electron microscopy(TEM). These methods were developed to improve and extend the information obtainable from TEM images of particulate samples, typically proteins or other large biological entities such as viruses. Individual images of stained or unstained particles are very noisy, and so hard to interpret. Combining several digitized images of similar particles together gives an image with stronger and more easily interpretable features. An extension of this technique uses single particle methods to build up a three dimensional reconstruction of the particle.

New methods of acoustic imaging are presented that provide images with reduced reverberation noise and images of nonlinear scattering and propagation parameters of the object, and estimation methods of corrections for wave front aberrations produced by spatial variations in the acoustic propagation velocity. The methods find use in a variety of applications such as acoustic imaging of geological structures, SONAR imaging of sub-sea objects, and medical ultrasound imaging. The methods are based on processing of the received signal from transmitted dual frequency band acoustic pulse complexes with overlapping high and low frequency pulses. The high frequency pulse is used for the image reconstruction and the low frequency pulse is used to manipulate the nonlinear scattering and/or propagation properties of the high frequency pulse.

We propose a finite-element method(FEM) deformable breast model that does not require elastic breast data for nonrigid PET/MRI [positron emission tomography/magnetic resonance imaging] breast image registration. The model is applicable only if the stress conditions in the imaged breast are virtually the same in PET and MRI. Under these conditions, the observed intermodality displacements are solely due to the imaging/reconstruction process. Similar stress conditions are assured by use of an MRI breast-antenna replica for breast support during PET, and use of the same positioning.

The interest of a ground based Slit Aperture Telescope(SAT) which operates with a long thin pupil has been underlined for applications in the field of high angular resolution observations by speckle-interferometric techniques. The SAT lies halfway between the Michelson stellar interferometer and the standard telescope with circular aperture. The authors present the principle of an image processing algorithm which could be used associated with a space-SAT for astronomical image reconstruction. A full coverage of the two dimensional Fourier plane of the astronomical object can be obtained by rotating the SAT around its optical axis. The method of image reconstruction is analogous to those used in computerized tomography and uses Fast Fourier Transform algorithm. The interest of the use of a rotating space-SAT among synthetic apertures is its capability of giving a reconstructed image of the astronomical object simultaneously in several light wavelengths, each colored image being comparable with the others for fruitful astrophysical applications.
