Confocal microscopy. Unlike "normal" microscopy, only a spot of a minimum size in the specimen is illuminated in confocal microscopy. To enable an image to be formed from this, the specimen must be scanned. The image of the spot is directed through a pinhole stop in an intermediate image plane. As a result, only light from the focal plane can reach the detector(a photomultiplier). All other(out-of-focus) planes are blocked out. This results in an "optical section". The images are stored electronically and displayed on a monitor. A series of optical sections can be recorded by moving a motor a slight distance along the z-axis each time an image has been recorded, after which the next image is then also recorded. Such a z-series permits the electronic reconstruction of the three-dimensional structure using suitable computer programs. The procedure, which is restricted to incident-light techniques, has completely revolutionized fluorescence microscopy in biology in particular.

L'utilisation de traceurs fluorescents très spécifiques permet de localiser in situ avec une résolution spatiale de l'ordre de 400 - 500 nanomètres, des composants macro-moléculaires tels que acides nucléiques et protéines, mais aussi de petites molécules telles que des peptides, lipides et même des ions.

La microscopie confocale permet la localisation in situ d'une sonde fluorescente dans des cellules d'un spécimen d'une épaisseur de 10 mm à 50 mm selon certains auteurs (5 mm à 30 mm plus raisonnablement), sans sectionnement mécanique préalable. SIAM is a technology that defies the current laws of imaging. It permits scientists to view images much smaller than the wavelength of light and approaching the atomic level, an achievement that has long been considered impossible according to the basic principles of classical optics. SIAM works by shining light through a thin sample onto an ultra-sharp, vibrating tip on the other side. The light scattering from the tip gets altered as the tip approaches within a nanometer from the sample surface, as it vibrates, due to the optical interaction with the local properties of the sample. An image is built up by recording the scattered light as the probe is scanned line by line. The scientists recorded the variations in the scattered light using a very sensitive device--an optical interferometer--which is capable of measuring modulations in light level corresponding to single photons. As the tip moved across the surface, these variations were mapped by a computer into a picture of the surface, showing features such as oil droplets and steps on mica that were only a few atoms in size.

Après s'être réfléchi sur l'échantillon et sur la lamelle, le faisceau d'analyse se combine avec le faisceau de référence dans un dispositif interférométrique. Un nouveau faisceau permet d'analyser les modifications des vibrations de la lamelle; il renseigne donc sur le relief de l'échantillon observé. En promenant ce dernier sous la micropointe, on peut reconstruire son image point par point avec une extrême précision.

Le signal reçu peut être ainsi organisé dans une matrice bi-dimensionnelle, où les paramètres de temps et de distance sont représentés respectivement en ligne par le cadencement des impulsions (PRF) et en colonne par la position des échos sur la ligne, une ligne étant acquise à chaque émission d'une impulsion radar (ou pulse). Une colonne de l'image représentera l'ensemble des réponses des réflecteurs placés à une distance donnée de l'antenne (distance radiale). Sa position sur la ligne est linéairement liée au temps mis par l'onde radar à lui revenir après s'être réfléchie sur le point cible, le radar étant considéré immobile durant ce temps.

The Gamma filter performs gamma map filtering. Gamma filtering is primarily used on radar data to remove high frequency noise(speckle) while preserving high frequency features(edges). The GAMMA filter performs spatial filtering on each individual pixel in an image using the grey level values in a square window surrounding each pixel. The dimensions of the filter must be odd, and can be from 3x3 to 11x11 pixels.

The frame size on 35 mm perforated film of an area 17. 5 x 23 mm. This is the standard 35 mm silent motion-picture frame size, and is also used for horizontal viewing as the frame size for filmstrips. Filmstrips are usually produced with special cameras. The same size image is also produced by normal half-frame cameras using standard 35 mm film, and producing twice the normal number of exposures per roll(i. e., 40) on a standard 20-exposure roll. As a comparison, the normal frame size(projected area of the film) for a 35 mm camera using perforated film is 23 x 34 mm, and is called double-frame size.
