Immobilization can result in significant changes in the properties of enzymes in comparison with the properties of the soluble enzyme. It is likely that the properties of the immobilized enzyme are nearer to the in vivo properties since dilute soluble enzyme studies normally ignore the complex realities of the intracellular environment. An immobilized enzyme system is the material which results from immobilization of an enzyme onto a support matrix. Magnetic support matrices and immobilized enzyme systems offer several advantages when compared to nonmagnetic systems. For example, separation of such materials from other nonmagnetic solids by use of a magnetic field can permit separations otherwise difficult or impossible to perform. Such a situation is represented by the case where two enzymes, with different deactivation times, are used concurrently on a substrate, with one immobilized enzyme system magnetic and the other nonmagnetic. The selection of conditions for immobilization is more difficult than in the cases of the other carrier binding methods described, and frequently involves more complicated and less mild conditions than the other methods, but, since covalent bonds are being formed, stable immobilized enzyme preparations which do not leach enzyme into solution in the presence of substrate or high ionic concentration solutions are formed in almost all cases.
