Immune checkpoints are a normal part of the immune system. Their role is to prevent an immune response from being so strong that it destroys healthy cells in the body. In another application of gene therapy we have used an adenoviral vector that carries the gene for herpes simplex virus thymidine kinase(HSV-tk). Cells expressing HSV-tk will convert the anti-viral drug ganciclovir(GCV) to a form that is toxic to rapidly dividing cells. Using this adenoviral vector we have repeatedly shown robust tumoricidal effects in a variety of animal models of CNS tumor and will soon take this treatment to phase I clinical trials. A subpopulation of lymphoid stem cells differentiates along T-cell lineage to become pre-T cells. These migrate to the thymus cell, where they become thymocytes then acquire different cell-surface antigen markers and functionally differentiate into subpopulations of T helper(inducer) cells, T suppressor cells, and T cytotoxic(killer) cells.
