Membrane filtration is the method of choice for the analysis of fecal coliforms in water. Samples to be tested are passed through a filter of particular pore size(generally 0. 45 micrometre). The microorganisms present in the water remain on the filter surface. When the filter is placed in a sterile Petri dish and saturated with an appropriate medium, growth of the desired organisms is encouraged, while that of other organisms is suppressed.... A 100 ml volume of a water sample is drawn through a membrane filter(0. 45 µm pore size) through the use of a vacuum pump. The filter is placed on a Petri dish containing M-FC agar and incubated for 24 hours at 44. 5 °C(112. 1 degrees F). This elevated temperature heat shocks non-fecal bacteria and suppresses their growth. 9, record 2, English, - membrane%20fecal%20coliform%20agar

2, record 2, French, - g%C3%A9lose%20pour%20d%C3%A9tection%20de%20coliformes%20f%C3%A9caux%20par%20filtration%20sur%20membrane

The Bartels PRIMA SystemTM Cytomegalovirus IgM Enzyme Immunoassay is a rapid, qualitative method for the detection of IgM Antibody to cytomegalovirus in human serum. Patient serum is diluted with specimen diluent that contains an absorbent to remove IgG. The absorbent prevents IgG/rheumatoid factor complexes from forming and prevents interfering competition from IgG during the test procedure. The treated serum is incubated with purified CMV antigens bound to EIA plate wells. If antibodies to CMV are present, they bind to the antigen and do not rinse off. Subsequently, when enzyme labeled anti-human IgM is added to the reaction site, it binds to the immobilized antibodies. After washing and the addition of a chromogenic substrate and stop solution, specimens containing antibodies to CMV produce a color endpoint reaction which is measured and compared to negative and positive controls, and may be measured spectrophotometrically. 1, record 12, English, - chromogenic%20substrate

A zone of inhibition surrounding the disk, as evidenced by a clear area in the otherwise confluent bacterial growth, indicates that growth of the microorganism was inhibited by the chemical on the disk.... With the proper controls the disk sensitivity method is a highly satisfactory technique for determining sensitivity of microorganisms to chemical agents. 1, record 14, English, - disk%20sensitivity%20method

The fermentation of solid substrates such as starchy or cellulosic materials can be approached in two different ways. The solid material can be suspended in an aqueous medium. A general problem with such preparations is that gelatinization of the starches, etc. can generate highly viscous broths causing poor mixing and O2 transfer at dry weight concentrations higher than 2% w/v. There is also the obvious diseconomy of adding so much water to the substrate only to remove it from the product. Alternatively, then, the problem is approached through ’solid state fermentation’, the growth of microorganisms on solid materials without the presence of free liquid. 3, record 25, English, - solid%20substrate%20fermentation
Record number: 25, Textual support number: 1 OBS
Such techniques can be divided into two basic types : thin layer and deep bed. In the former system, the organisms are grown on the surface of the substrate, which is spread on shallow trays about 2-4 cm deep; the trays are incubated in air-conditioned rooms or cabinets. The simplest deep bed systems consist of heaps of substrate that may be stirred periodically by hand. More sophisticated, fully automated systems include rotating drums, conveyor belt systems, which pass through air-conditioned tunnels, or deep tanks through which air is blown. The substrates used are generally based on wheat bran. 1, record 25, English, - solid%20substrate%20fermentation

A method used to detect and enumerate antibody producing cells. Cells obtained from, e. g. the spleen of an animal that has been immunized against sheep erythrocytes, are mixed into molten Agarose containing sheep erythrocytes. The agarose is poured into a Petri dish, allowed to set and then incubated at 37 ° C. Later, complement is poured into the dish, and any cells that have secreted antibody are revealed by the appearance of a zone of haemolysis round each of them. The agarose layer can be dried and stained so that the cells may be identified. The technique described reveals IgM secreting cells forming direct plaques. By the addition of an anti-IgG serum before the complement, indirect plaques due to IgG secreting cells can be revealed. Cells producing antibodies to antigens other than erythrocytes can be detected by coating the rythrocytes with the antigen as in passive haemolysis. 1, record 27, English, - balmolytic%20plaque%20test

[Molecular cloning] is likely to offer unique possibilities for investigating mechanisms of gene regulation in higher organisms. For example, a bacterial culture with incorporated eukariotic DNA could be incubated with components to be tested for regulatory effects such as enzymes or repressors. Their influence on gene expression could be investigated. 1, record 28, English, - gene%20regulation
