Tetanus(lockjaw) is caused by neurotoxins produced by the bacteria Clostridium tetani, particularly in deep or closed wounds, such as puncture wounds, but may occur, due to intestinal infections. This highly fatal disease is a constant threat for all horses as the bacteria are widely distributed in soil and manure.... Clinical signs of tetanus in affected horses begin following an incubation period of 3 days to several weeks, but usually 1 to 2 weeks. Muscle spasms and stiff paralysis progressing from the head to the neck, front, and then hind legs occurs.... The horse may be unable to swallow, resulting in saliva dripping from the mouth and regurgitation of ingested food and water through the nose. Convulsions, muscle rigidity, sweating, and fever may follow.

Les spasmes commencent généralement au niveau de la tête et du cou (hyperextension de l'encolure, procidence de la 3e paupière, lèvres et nez retroussés), puis évoluent en une raideur généralisée, une incapacité à se nourrir, un décubitus et la mort par arrêt respiratoire.

No virus has ever been causally associated with CJD [Creutzfeldt-Jacob disease]. If such a virus exists its properties are very unusual. It must be ubiquitous but normally non pathogenic. PrP [prion protein] would be essential for its infectivity e. g. perhaps serving as a receptor. Prion strains exist(more than 20 strains have been identified in mice infected with scrapie prions and several strains from humans). These strains breed true in inbred mice and differ in their incubation times and lesion profiles. This observation and the reluctance to accept because of precedence the existence of a replicating agent lacking a nucleic acid genome has led to the virino hypothesis. It proposes that a small nucleic acid in association eith PrP is the infectious agent. It is not fully consistent with all of the data and no such nucleic acid sequence has ever been detected.

The affinity label, N-tosyl-L-phenylalanylchloromethyl ketone(TPCK), was designed to resemble a normal substrate for chymotrypsin. However, instead of the usual susceptible structure of the substrate(-CO-NH-), TPCK contains the-CO-CH2Cl group, a potent alkylating agent. Incubation of TPCK with chymotrypsin causes it to bind to the active site in the same way as the normal substrate. However, instead of hydrolysis of the substrate, alkylation of the essential His 57 residue occurs, indicating that the latter is close to the susceptible bond normally undergoing hydrolysis. The alkylated His residue can be isolated following complete hydrolysis of all the peptide bonds of the enzyme.
