The division of the nucleotide sequence of a nucleic acid into consecutive three-nucleotide codons.

OBS

There are three different frames(starting a codon with the first, second, or third nucleotide). Generally, only one frame encodes a polypeptide; this frame is specified by the location of the initiation codon of the polypeptide. The deletion or addition of a nucleotide throws off the frame after that point, making non-sense of the remaining code; this is called a frameshift mutation.

A codon that codes for the first amino acid in a polypeptide sequence.

CONT

A special transfer RNA (tRNA) recognizes this codon (AUG, or rarely, GUG) and starts the chain with the amino acid methionine (met) or rarely, valine, in the cytosol of eukaryotes or with N-formyl methionine (F-met) in prokaryotes, mitochondria, and chloroplasts. A different tRNA reads AUG as met in the middle of a chain.

A baculovirus transfer vector was constructed using a cloned copy of the autography californica nuclear polyhedrosis virus(AcNPV) polyhedrin gene. The ATG translation initiation codon and 32 bases of the polyhedrin mRNA 5’ leader sequences were deleted and replaced with a cDNA clone of the A/PR/8/34 influenza virus haemagglutinin(HA) gene. The hybrid HA/polyhedrin gene was transferred to AcNPV and polyhedrin-negative recombinants identified which expressed HA activity.
