Chez les procaryotes, la région du promoteur comporte environ 40 paires de bases, comprenant quelques bases hautement spécifiques situées entre 35 et 30 bases et entre 12 et 7 bases avant le début de la transcription. Chez les eucaryotes, la région du promoteur comporte plusieurs centaines de paires de bases précédant le début de la transcription, et on y trouve, pour presque tous les gènes, des séquences caractéristiques : boîte TATA, boîte CCAAT, boîte GC. Ces boîtes fixentspécifiquement certaines protéines appelées facteurs transrégulateurs. La boîte TATA, située à environ 25 bases du point de départ de la transcription, sertapparemment à fixer le complexe multiprotéique nécessaire à l'initiation de la transcription par la RNA-polymérase. D'autres séquences du promoteurinterviennent dans la régulation de la transcription, séquences dites éléments cis-régulateurs.

The transcribing enzyme, RNA polymerase, binds to a promoter region that(in the bacterium Escherichia coli) has the specific sequences shown(or minor variation of them) about 10 base pairs and 35 base pairs before a transcription initiation site; beyond the end of the structural gene a termination region causes the polymerase to cease transcription.
