In another application of gene therapy we have used an adenoviral vector that carries the gene for herpes simplex virus thymidine kinase(HSV-tk). Cells expressing HSV-tk will convert the anti-viral drug ganciclovir(GCV) to a form that is toxic to rapidly dividing cells. Using this adenoviral vector we have repeatedly shown robust tumoricidal effects in a variety of animal models of CNS tumor and will soon take this treatment to phase I clinical trials. The protein kinase PKR is a component of the interferon-induced anti-viral defense mechanism in mammalian cells. Upon activation by binding double-stranded RNA in infected cells, the kinase down-regulates the cellular translational apparatus, and thus impairs viral protein expression. To overcome the inhibitory effects of PKR, viruses have developed efficient methods to prevent the activation or function of the kinase. Activation of macrophages by bacterial lipopolysaccharide(LPS) induces transcription of genes that encode for proinflammatory regulators of the immune response. Insoluble peptidoglycan (PGN) from staphylococcal cell walls as well as soluble PGN (sPGN) secreted by staphylococci in the presence of beta-lactam antibiotics induced TNF- alpha mRNA and secretion of bioactive TNF-alpha in the murine RAW264.7 macrophage cell line.
