DNase Agar. DM132 A medium for the presumptive identification of pathogenic staphylococci by demonstration of DNase production.

Deoxyribonuclease(DNAse) agar actually has DNA in it. Deoxyribonuclease is an exoenzyme excreted from the cell which will break the DNA down into smaller molecules. There are 2 kinds of DNAse agar that can be used. 1) The first medium has no indicator, but is does require HCl(hydrochloric acid) reagent to be added after growth has occurred on the plate. Without DNAse enzyme, the HCl precipitates out the undestroyed DNA. The rest of the agar with the remaining DNA, which binds to HCl will have turned cloudy. Around the growth area, if the bacterium has made DNAse and destroyed the DNA, there will be clear zones seen. 2) DNAse agar with the indicator methyl green can be used. In this medium, when the DNA in the medium is hydrolyzed by the bacterial DNAse enzyme, the pieces of cut DNA release from binding to methyl green, and you will see a yellowish/orange zone around the growth area. OBJECTIVE : To identify the presence of the DNAse enzyme.

An azo dye used as an acid-base indicator; used for titrations involving weak bases.

Methyl orange is an intensely colored compound used in dyeing and printing textiles. It is also known as C. I. Acid Orange 52, C. I. 13025, helianthine B, Orange III, Gold orange, and Tropaeolin D. Chemists use methyl orange as an indicator in the titration of weak bases with strong acids. It changes from red(at pH 3. 1) to orange-yellow(at pH 4. 4).... Methyl orange is synthesized by reaction of sulfanilic acid, sodium nitrite, and dimethylaniline.

Methyl Red Voges Proskauer Test(MR-VP)-differentiates members of family Enterobacteriaciae based on ability to produce acid end products from fermentation of glucose. Enterobacteriaciae divided into two groups based on type of formic acid fermentation performed. 1.) Mixed acid type formic acid fermentation-results in production of ethanol and lactic, acetic, succinic, and formic acids as fermentation end products. If mixed acid fermentor, then when Methyl Red added to inoculated broth, medium turns red. If butanediol fermentor, then the color of the medium will change to yellow. Butanediol fermentor yields orange/yellow color after addition methyl red. 2.) Butanediol fermentors produce acidic fermentation products.
