Research is on hairpin catalytic RNA, a catalytic RNA discovered and characterized here in my laboratory at Northern Illinois University. Hairpin catalytic RNA is a molecule which can be engineered to very efficiently cleave target RNA sequences in a unique and efficient reaction. In the March 28, 1997 issue of Science, Li et al describe a protein which is mutated in a number of human cancers, including those from breast, brain and prostate. This protein interacts with actin filaments and is a putative protein tyrosine phosphatase. Li et al have called this protein PTEN(phosphatase and tensin homolog deleted on chromosome 10). Steck et al, who simulatenously located the same gene have adopted the name, MMAC1(mutated in multiple advance cancers 1). The 5. 5-kb mRNA of the PTEN/MMAC1 is widely expressed in human tissues and it appears to have alternately spliced variants. The N-terminal domain of the gene codes for a 403-amino acid protein that is likely a protein phosphatase and is related to the CDC14 which is an enzyme that controls the cell division.

PGN and sPGN also induced rapid and dose-dependent tyrosine phosphorylation of several cellular proteins, including lyn and mitogen-activated protein kinases(extracellular signal-regulated kinases; but not hck, fgr, or vav) and increased the activities of mitogen-activated protein and rsk kinases. These PGN-and sPGN-induced effects were qualitatively similar to the effects induced by ReLPS, but higher concentrations of PGN and sPGN than ReLPS were required.

Tomato ringspot nepovirus (TomRSV) infects small fruits and tree fruits. Coat and movement proteins of TomRSV are expressed coordinately via proteolytic processing of a polyprotein precursor at specific cleavage sites by a virus-encoded protease. Therefore the two cleavage sites involved in the release of the coat and movement proteins from the precursor polyprotein must be recognized at equal efficiency by the TomSVP protease. These results provide important information toward the inhibitors of viral proteases, which has potential as a novel strategy for engineering resistance to plant viruses.

A cDNA library was prepared from human leukocytes. Interferon represents less than 0. 1 percent of the total mRNA in these cells. The cDNA library was broken down into eight groups, and total plasmid DNA was prepared from each group. The presence of interferon cDNA in each group was determined by using the hybrid-selection procedure. When an interferon group of clones was identified, it was subdivided into groups containing fewer clones, and the process was repeated. Four such subdivisions were necessary to obtain a pure cDNA clone for human interferon.
