Reduction of β-cell mass and function is central to the pathogenesis of type 2 diabetes. The terms glucotoxicity, lipotoxicity, and glucolipotoxicity are used to describe potentially responsible processes. The premise is that chronically elevated glucose levels are toxic to β-cells, that elevated lipid levels in the form of circulating free fatty acids(FFA) also have toxic effects, and that the combination of the two, glucolipotoxicity, is particularly harmful.

Paradoxically, because of its toxicity and its high specificity for beta cells, streptozotocin has been used with some success to treat a rare tumor of human beta cells known as insulinoma. In general, however, drugs and chemicals that kill beta cells were employed merely as tools for investigating the pathogenesis of diabetes in experimental animals. Then in 1975 a rodent poison known as Vacor, which has a molecular structure resembling that of streptozotocin, was introduced into the U. S.

Ewing's sarcoma [a malignant tumor of the bone marrow] is classified in a group of round cell lesions because of similar histologic and radiographic features. Lymphoma is another member of this group. Ewing's sarcoma tends to occur in children and young adults, whereas primary lymphoma of bone(usually non-Hodgkin's type) is seen in older individuals. The two entities may be difficult to differentiate. Ewing's sarcoma is of unknown pathogenesis and is highly malignant.

Latency-Lentiviruses do not show true latency(unlike Herpes viruses or lambda) but do have the capacity to control the expression of their genome by means of virus-encoded trans-acting regulatory proteins(tat and rev). Antigenic variation-new variants continually arise. In other Lentiviruses such as CAEV, each new antigenic variant results in a flare up of disease.

Interpretation of changes in bone marrow during infectious processes is quite complex. This paper reports bone marrow lesions observed in pigs inoculated with a moderately virulent ASF virus strain and studies their relationship to the pathogenesis of the disease. In this work, we have carried out the structural and ultra-structural study of the bone marrow of 14 Large White x Landrace pigs that were inoculatd by the intramuscular route with 105 50% hemodsorbing doses(HAD50) of the Dominican Republic' 78 ASF virus strain. The inoculated pigs were killed at 3, 5, 7, 9, 11, 13, 15 and 17 days postinjection.

Osteonecrosis, without evidence of an inflammatory process or sepsis, is usually regarded as secondary to loss of the blood supply(avascular necrosis of bone). Some argue that metabolic changes in the osteocytes are responsible for bone death in certain forms of the disease and recommend the general term osteonecrosis. An older synonym that emphasizes the lack of an inflammatory component is aseptic necrosis.
