DEF

Type of chromatography developed following the observation that certain proteins were unexpectedly retained on affinity gels containing hydrophobic spacer arms. This concept was extended, and families of hydrophobic adsorbents prepared using a homologous series of hydrocarbon chains of different lengths. In practice most proteins can be satisfactorily purified using C-8 or phenyl substituents. Hydrophobic interactions are strongest at high ionic strength, so adsorption to a hydrophobic matrix can be conveniently performed after a salt precipitation or ion exchange chromatography step. Elution can be effected by altering the solvent ionic strength, pH, or composition or by the use of a dielectric constant modifier such as ethanediol.

CONT

When carrying out the hydrophobic chromatography of human urokinase..., the solution containing the crude urokinase which is prepared from human urine in the usual simple method, is adjusted to a salt concentration of 0. 2 to 2 M, preferably 0. 2 to 1 M. The solution is then made to flow through the hydrophobic column packed with the protein adsorbent of this process.... The pH is not critical when carrying out the hydrophobic chromatography, however, the pH is usually adjusted to pH 5. 5 to 10, preferably 6 to 8. 5 at which pH urokinase is stable against inactivation. Purified urokinase can be obtained by treating the effluent from the hydrophobic column in a usual method, namely by lyophilization after desalting.

CONT

DEF

Quotient of the luminous flux incident on an infinitesimal element of a surface containing the point under consideration, by the area of that element.

CONT

Illuminance (usually ’E’ in formulas) is the total amount of visible light illuminating (incident upon) a point on a surface from all directions above the surface. This "surface" can be a physical surface or an imaginary plane. Therefore illuminance is equivalent to irradiance weighted with the response curve of the human eye.

OBS

Standard unit for illuminance is Lux(lx) which is lumens per square meter(lm/). There are several older units of illuminance : footcandle : 1 fc=10. 764 lx; phot : 1 ph=10 000 lx. Typical illuminance values are : 1 lx=full moon; 10 lx=street lighting; 100-1 000 lx=workspace lighting; 10 000 lx=surgery lighting; 100 000 lx=plain sunshine.

OBS

"Illumination" is a deprecated term for "illuminance".

OBS

illuminance: term officially approved by the RADARSAT-2 Terminology Approval Group (RTAG).

To complete the definition of pH, values of pH have been assigned to standard solutions. The National Bureau of Standards has recommended five solutions as primary standards. CONT

DEF

Standard that is designated or widely acknowledged as having the highest metrological qualities and whose value is accepted without reference to other standards of the same quantity.

CONT

A standard solution is prepared by dissolving an accurately weighed quantity of a highly pure material called a primary standard and diluting to an accurately known volume in a volumetric flask.

CONT

To complete the definition of pH, values of pH8, have been assigned to standard solution. The National Bureau of Standards has recommended five solutions as primary standards. One of these, 0. 05 M potassium hydrogen phthalate, has been adopted as the primary standard of pH in Great Britain and Japan.

OBS

The concept of primary standard is equally valid for base quantities and derived quantities.

DEF

OBS
