STED [stimulated emission depletion] microscopy was the first high-resolution microscopy concept applied more widely for the study of biological questions. As performed in classical confocal laser scanning microscopy, a conventional laser beam is used to stimulate the fluorescent molecules in what is a normal excited spot(of several hundred nanometers in diameter). A second toroidal(doughnut shaped) beam is used to quench the fluorophores by stimulated emission. By overlapping the two beams, fluorescence is allowed only from the center of the spot, which thus generates a much smaller exciting focal spot... Thus, in practical terms, a smaller effective PSF [point spread function] is generated that causes less blurring and, therefore, results in higher resolution. The nanomotor has a positioning stroke up to some centimeters while operating with atomic resolution and thus builds a bridge over eight orders of magnitude. The nanomotor is a piezo driven linear motor consisting of a cylindrical housing and a slider with a free axial hole inside. It utilizes a piezo tube for fine positioning and a pulse wave produced by the same piezo tube for coarse movements. Every point of its stroke can be reached with a speed of up to 5 mm/s. In its free axial hole the nanomotor can transport a tip, a hose, an electrode, a hypodermic needle, a glass fiber or even a microgripper. The nanomotor is available in three different versions. The "small" nanomotor is half the size of a match stick. It can lift six times of its own mass. The dynamic force of this nanomotor is so high, that even imprints in diamond surfaces can be produced. A confocal laser scanning microscope is a light microscope having a dramatically higher axial resolution than a conventional microscope. In other words, a confocal microscope is capable of optical sectioning. This is accomplished by illuminating the specimen one point at a time with a focused laser beam and using an optical arrangement, where only the light originating from the focus can be detected. The image is formed by scanning the laser beam over the specimen and recording the image into computer memory. Le microscope confocal à balayage laser (MCBL) est un microscope optique à éclairage épiscopique géré par un ordinateur qui est capable de sectionner optiquement un échantillon épais. Son image est ensuite reconstruite en superposant toutes ou certaines des sections optiques. Seuls les rayons image venant du plan focal, qui est confocal avec le plan du diaphragme, parviennent au détecteur. Les autres rayons, provenant d'autres plans focaux, sont bloqués par le diaphragme [...]. L'image résultante obtenue par balayage au niveau du plan focal a une épaisseur extrêmement limitée (section optique) et n'est pas perturbée par des images floues des plans situés au dessus ou au dessous. Comme l'ordinateur déplace la platine dans la direction de l'axe optique Z avec des mouvements micrométriques prédéfinis après chaque balayage, le MCBL ne peut pas recueillir les images provenant d'autres plans. L'ordinateur stocke les images correspondants aux sections optiques successives.
