Computational sequence analysis, that is, prediction of local sequence properties, homologs, spatial structure and function from the sequence of a protein, offers an efficient way to obtain needed information about proteins under study. Since reliable prediction is usually based on the consensus of many computer programs, meta-servers have been developed to fit such needs. 1, record 1, English, - computational%20sequence%20analysis

The ability of certain restriction enzymes to recognize rather short, specific sequences of DNA as targets for cleavage opens a direct approach to gene structure. It is possible to proceed to determine the sequence of DNA between points that are sufficiently close together by suitable choice of points, regions can be connected into a sequence of the entire gene. The sequence of the DNA then can be compared with the sequence of the protein that the gene represents. This allows the regions corresponding to the protein to be delineated; and, by extending the sequence in either direction, the distance to the next gene can be determined. 2, record 5, English, - gene%20structure

Janet started from the fact that the series of chemical elements is a continuous sequence, which he represented as a helix traced on the surfaces of four nested cylinders. In the absence of a suitable crystal structure, computer methods must predict protein conformation from sequence. The most powerful approaches for structure prediction are based on sequence homology or a more general analogy with known crystal structures. Improvements in these methods will require access to data bases containing the basic motifs of protein architecture... The database will also enable us to extract and store tertiary motifs... Optimal application of protein engineering technology will require understanding how the amino acid sequence of a polypeptide chain specifies its spatial conformation. The critical amino acid instructions directing these pathways appear to be dispersed through the sequence... Recombinant DNA(rDNA) molecules are DNA sequences that result from the use of laboratory methods(molecular cloning) to bring together genetic material from multiple sources, creating sequences that would not otherwise be found in biological organisms. Recombinant DNA is possible because DNA molecules from all organisms share the same chemical structure; they differ only in the sequence of nucleotides within that identical overall structure. Therefore, when DNA from a foreign source is linked to host sequences that can drive DNA replication and then introduced into a host organism, the foreign DNA is replicated along with the host DNA. 2, record 12, English, - recombinant%20DNA

The protein Sonic Hedgehog (Shh) controls patterning growth during vertebrate development. Here we demonstrate that it binds Patched (vPtc), which has been identified as a tumour-suppressor protein in basal cell carcinoma, with high affinity. We show that Ptc can form a physical complex with w newly cloned vertebrate homologue of the Drosophila protein Smoothened (vSmo), and that vSmo is coexpressed with vPtc in many tissues but does not bind Shh directly. 4, record 16, English, - Smoothened

2, record 24, French, - tomodensitom%C3%A9trie

The lipocalin protein family is a large group of small extracellular proteins. The family demonstrates great diversity at the sequence level; however, most lipocalins share three characteristic conserved sequence motifs, the kernel lipocalins, while a group of more divergent family members, the outlier lipocalins, share only one.

The principle factor responsible for the transcriptional state of the c-fos proto-oncogene is the serum response factor(SRF) which binds a conserved DNA sequence called serum response element(SRE) present in the promoter region of the c-fos gene. In order to elucidate the molecular basis of this interaction, we have solved the crystal structure of a recombinant SRF-SRE complex(Nature, 376 : 490-298, 1995). 3, record 36, English, - serum%20response%20factor

Several proteins or parts of proteins that do not have homologous sequences are observed to adopt a similar tertiary arrangement of packed [alpha]-helices and [beta]-strands.... Generally, it is an open question whether the similarity of tertiary structures arises by convergent evolution to a structural and/or functional favourable conformation or by divergence during which sequence homologies have been lost.... Combined with the search for key amino acid residues, it is likely that structural similarity will be recognized by comparing profiles of secondary structure predictions and by using empirical rules governing the packing of [alpha]-helices and [beta]-strands. 1, record 46, English, - beta%2Dstrand

3, record 46, French, - structure%20b%C3%AAta

A new method for direct sequence analysis of DNA restriction fragments uses synthetic covalently linked complementary oligodeoxynucleotides, as universal sequencing primer linkers(splinkers). These splinkers were designed to contain an inverted repeat sequence which forms a double-stranded hairpin structure with a known restriction site. The splinkers were characterized by their ability to be self-ligating(dimerized) and by their restriction digest product analysis of both the monomer and dimer. 1, record 48, English, - universal%20sequencing%20primer%20linker
