Transcription activator-like effector nucleases (TALENs) are restriction enzymes that bind to DNA based on its specificity and can be tailored to cut sequences of DNA. They are made by combining TAL [transcription activator-like] binding domains, which allows entry into a cell, and a DNA cleavage domain, which is helpful in gene editing. These can be tailored to bind to any targeted area in the genome to cut any desired specific location. 5, record 1, English, - transcription%20activator%2Dlike%20effector%20nuclease

Base editing.... It enables precise changes to individual nucleotides within the genome without creating double-strand breaks.... base editing directly converts one DNA base to another at a specific target site. [Base editing] allows for precise single-base changes, such as converting a C-G base pair to a T-A base pair using a cytosine base editor(CBE) or an A-T base pair to a G-C base pair using an adenine base editor(ABE), without introducing insertions or deletions. 2, record 2, English, - base%20editing

Guide RNAs (gRNAs) are tiny RNA [ribonucleic acid] transcripts that guide the editing machinery through base-pairing with the mRNAs [messenger ribonucleic acids] in edited regions. gRNAs are complementary to small segments of the fully edited mRNAs, and thus serve as mini-templates to guide the addition and deletion of Us from the pre-edited mRNA as they form a locally double-stranded RNA helix. 4, record 3, English, - guide%20ribonucleic%20acid

No one studying molecular biology can escape the impact that the CRISPR/Cas9 system has had on the field. In its natural environment, it functions as an adaptive bacterial immune system that can remember past viral infections. However, it is mainly known and popular as a highly versatile and precise tool for gene editing.... At the heart of the system is Cas9, a programmable and sequence‐specific endonuclease that introduces double‐strand breaks into DNA, which can be exploited to modify the DNA sequence in many different ways. The Cas9 cleavage site is determined by a small RNA molecule, termed guide RNA(gRNA), that base pairs with the target DNA and at the same time binds to Cas9 to initiate the site‐specific double‐strand break... 5, record 3, English, - guide%20ribonucleic%20acid

Site-directed mutagenesis technique based on the use of an oligodeoxynucleotide cassette(a synthetic double-stranded DNA fragment). The strategy involves the introduction of unique silent restriction sites designed to closely flank the target codon. Digestion with appropriate restriction enzymes produce a gap that can be filled with a duplex synthetic oligonucleotide cassette. The cassette is designed to restore the coding sequence in the gap and to introduce an altered target codon. 2, record 12, English, - cassette%20mutagenesis

1, record 12, French, - mutag%C3%A9n%C3%A8se%20par%20cassette

In a process called "molecular combing", DNA molecules attached at one end to a solid surface were extended and aligned by a receding air-water interface and left to dry on the surface. Molecular combing was observed to extend the length of the bacteriophage Lambda DNA molecule to 21.5+/-0.5 micrometers (unextended length, 16.2 micrometers). 1, record 13, English, - molecular%20combing

With the combing process, it was possible to :(i) extend a chromosomal Escherichia coli DNA fragment(10^6 base pairs) and(ii) detect a minute quantity of DNA(10^3 molecules). A very original set of experiments carried out by the Unit of Molecular Biology of Gene Expression concerns another protein essential for mouse development that is also a transcription factor. This factor known as KBF2 or RBP-Jk is homologous to a Drosophila gene product, Su(H)(Suppressor of Hairless), which belongs to the Notch signal transduction cascade involved in regulating the competence of a cell to respond to specific developmental cues. A likely function of the Su(H) protein is to serve as an anchor on DNA for non DNA-binding transcriptional activators. This function is conserved in the Notch pathway : the activation of this transmembrane receptor likely involves a proteolytic cleavage step which releases its intracellular domain; following nuclear translocation, it activates its target genes by binding to the Su(H) protein, which is already bound to several sites in the target promoters. 2, record 18, English, - Su%28H%29%20protein
