Guide RNAs (gRNAs) are tiny RNA [ribonucleic acid] transcripts that guide the editing machinery through base-pairing with the mRNAs [messenger ribonucleic acids] in edited regions. gRNAs are complementary to small segments of the fully edited mRNAs, and thus serve as mini-templates to guide the addition and deletion of Us from the pre-edited mRNA as they form a locally double-stranded RNA helix. 4, record 1, English, - guide%20ribonucleic%20acid

No one studying molecular biology can escape the impact that the CRISPR/Cas9 system has had on the field. In its natural environment, it functions as an adaptive bacterial immune system that can remember past viral infections. However, it is mainly known and popular as a highly versatile and precise tool for gene editing.... At the heart of the system is Cas9, a programmable and sequence‐specific endonuclease that introduces double‐strand breaks into DNA, which can be exploited to modify the DNA sequence in many different ways. The Cas9 cleavage site is determined by a small RNA molecule, termed guide RNA(gRNA), that base pairs with the target DNA and at the same time binds to Cas9 to initiate the site‐specific double‐strand break... 5, record 1, English, - guide%20ribonucleic%20acid

Research is on hairpin catalytic RNA, a catalytic RNA discovered and characterized here in my laboratory at Northern Illinois University. Hairpin catalytic RNA is a molecule which can be engineered to very efficiently cleave target RNA sequences in a unique and efficient reaction. The catalytic RNA can thus be used for the specific down regulation of gene expression since it can be designed to destroy mRNA. Current applications being developed are down regulation of selected genes, including those in the virus HIV-1. Thus it has potential as an AIDS therapeutic. 1, record 4, English, - catalytic%20RNA

For gene therapy to realize its clinical potential, there must be efficient and safe strategies of delivering therapeutic genes to somatic cells in vivo. Perhaps this problem simply represents a special case of drug delivery in which the therapeutic gene constitutes the drug. But the relevance of traditional drug development to technological developments in gene therapy is questionable. Recombinant genes that can independently express a therapeutic RNA or protein are extraordinarily large and polar molecules--10, 000 times larger than traditional pharmaceutical agents. Moreover, the administration of genes as therapy requires vehicles--so-called vectors--that encapsulate the gene and guide it to the target cell. The binding of the gene to the cell, its internalization, the transport of the administered genome to the nucleus, and the expression of the gene all constitute potential limitations of this process. 1, record 13, English, - therapeutic%20gene

1, record 13, French, - g%C3%A8ne%20%C3%A0%20vis%C3%A9e%20th%C3%A9rapeutique

A very original set of experiments carried out by the Unit of Molecular Biology of Gene Expression concerns another protein essential for mouse development that is also a transcription factor. This factor known as KBF2 or RBP-Jk is homologous to a Drosophila gene product, Su(H)(Suppressor of Hairless), which belongs to the Notch signal transduction cascade involved in regulating the competence of a cell to respond to specific developmental cues. A likely function of the Su(H) protein is to serve as an anchor on DNA for non DNA-binding transcriptional activators. This function is conserved in the Notch pathway : the activation of this transmembrane receptor likely involves a proteolytic cleavage step which releases its intracellular domain; following nuclear translocation, it activates its target genes by binding to the Su(H) protein, which is already bound to several sites in the target promoters. 2, record 14, English, - Su%28H%29%20protein

1, record 15, English, - therapeutic%20gene

1, record 16, French, - g%C3%A8ne%20cible

For now most scientists interested in gene augmentation are planning to remove cells from patients, introduce a therapeutic gene and return the altered cells to the subject. Some day, however, physicians may directly inject patients with genes linked to substances that will deliver those genes to specific target cells. 1, record 18, English, - gene%20augmentation
