For example, the concept of scaffold hopping, which could be considered a subset of the concept of bioisosterism..., might be applied during lead optimization to identify one or more new series of compounds having a fundamentally different core structure to improve, for example, its potency or selectivity profile, synthetic accessibility, intellectual property position, or pharmacokinetic properties. The goal of scaffold hopping is to attain novel structures by changing the central core framework(the scaffold) of active compounds without affecting its binding to the target receptor. 2, record 1, English, - scaffold%20hopping

deconvolution microscope : using the deconvolution microscope, researchers can see the locations of target molecules inside the cell. Seeing where glowing molecules reside within a cell is helpful for a range of research projects--from studies of how the distribution of various proteins changes during an organism's development to investigations of how administration of a drug alters the locations of certain proteins. The microscope, called DeltaVision, is manufactured by Applied Precision, based in Issaquah, Wash. It works by magnifying images and quantifying the intensity of light shed by the sample. The user specifies which colors of light are of interest. For instance, a user can program the microscope to report the intensity of a particular shade of green shining from the cytoplasm of a cell. After a researcher uses the microscope to make a series of optical slices through a specimen, the scope's computer "deconvolves" the images--in other words, it subtracts the out-of-focus light. Then the computer stacks together the images of the slices to reconstruct the original microscope sample. This gives researchers gh-resolution images of the sample's three-dimensional structure. 2, record 6, English, - wide%20field%20deconvolution%20microscopy

Nearly every drug molecule works through a structural interaction with a "target" or "receptor" molecule or protein that plays key roles in all biological processes. In the most common model for this interaction, the drug molecule inserts itself into a functionally important crevice of its target protein, like a key in a lock. The molecule then binds there and either induces or, more commonly, inhibits the protein's normal function. This universal drug-target scheme suggests a powerful alternative approach to drug discovery : if it were possible to identify, in advance, the appropriate protein target for a given therapeutic need and if enough were known about the distinguishing structure of that target protein, it ought to be feasible to design the structure of an ideal drug to interact with it. 2, record 14, English, - target%20protein

Improved uniformity of the laser for-al spot profile is a necessary criterion for the success of high intensity laser fusion and to improve the quality of data from moderate intensity laser driven shock experiments in solids. Recently developed laser smoothing techniques rely on both temporal smoothing and thermal smoothing to produce uniform ablation pressures on the target surface. Thermal smoothing is not effective during the low intensity portion of a laser pulse and the resultant shock structure mirrors the residual laser nonuniformities. The impact of the first(nonuniform) shock can be diminished by using foam layers, a precursor flash, shallow angles of incidence, and by adiabatically compressing the target with a temporally long, slowly rising laser pulse. 1, record 18, English, - thermal%20smoothing
