L'addition de bile ou de sels biliaires à une culture neutralisée active une autolysine, impliquée dans la division cellulaire, qui clive la liaison entre l'acide N-acétyl-muramique et l'alanine. Le peptidoglycane est ainsi altéré et la bactérie apparaît soluble dans la bile. En pratique, la recherche de la lyse par la bile est rarement effectuée car elle est de réalisation plus complexe que la recherche de la sensibilité à l'optochine.
