Site-directed mutagenesis technique based on the use of an oligodeoxynucleotide cassette (a synthetic double-stranded DNA fragment). The strategy involves the introduction of unique silent restriction sites designed to closely flank the target codon. Digestion with appropriate restriction enzymes produce a gap that can be filled with a duplex synthetic oligonucleotide cassette. Technique de mutagénèse dirigée in vitro qui consiste à créer dans le gène une "cassette" encadrée par deux sites d'enzyme de restriction. Il est ensuite aisé de remplacer cette cassette par une autre, constituée de deux brins d'ADN synthétique, pour introduire la mutation que l'on veut dans la zone de la cassette.
