Cette ADN polymérase de grande pureté se révéla incapable de produire, à partir d'une matrice d'ADN bactérien, un ADN biologiquement actif. Cet échec s'explique par le fait que l'ADN extrait d'une bactérie telle que B. subtilis constitue une matrice médiocre pour cette enzyme. Une bonne matrice serait, par exemple, le chromosome naturel ayant la forme d'une boucle à double brin d'environ un millimètre de circonférence.
