Les matrices hydrophiles résultent du mélange d'une ou de plusieurs substances actives avec un agent gélifiant. La libération se déroule en plusieurs étapes. Au contact de l'eau ou des fluides digestifs, une très petite fraction de la substance active se dissout immédiatement. Puis la pénétration progressive du solvant permet l'hydratation du système et la gélification des macromolécules, lesquelles forment une couche visqueuse de plus en plus épaisse à partir de laquelle la substance médicamenteuse est libérée après dissolution et diffusion dans le gel.
