La biomasse lignocellulosique subit d'abord un prétraitement physicochimique destiné à améliorer sa susceptibilité à l'hydrolyse enzymatique. L'hydrolyse enzymatique met en action les enzymes sur le substrat prétraité; après séparation du résidu non hydrolysé, les sucres produits sont transformés en solvants par la fermentation acétonobutylique; ces solvants sont ensuite distillés pour donner un mélange renfermant approximativement deux tiers de butanol, un tiers d'acétone et des traces d'éthanol.
