Technique de séquençage qui utilise des réactions chimiques interrompant spécifiquement la chaîne de DNA en position A, T, G ou C. Ainsi le DNA, marqué à son extrémité 5' par le 32P, est dégradé partiellement et spécifiquement au cours de quatre réactions séparées. L'électrophorèse de ces produits de dégradation sur les gels de polyacrylamide mis au point chez Sanger, suivie d'une autoradiographie, permet la lecture directe de la séquence de DNA.
