[Les] inhibiteurs nucléosidiques de la transcriptase inverse (INTI) [sont des] anti-rétroviraux [actifs] sur VIH-1 et VIH-2. [Ces] analogues de bases nucléiques nécessitent pour être actifs d'être phosphorylés dans le milieu intra-cellulaire. Ils rentrent ensuite en compétition avec les substrats naturels de la transcriptase inverse et inhibent l'action de cette dernière. Ils bloquent ainsi la fabrication d'ADN pro-viral.

Medicamento antirretroviral análogo de los nucleósidos cuya estructura química constituye una versión modificada de un nucleósido natural. Este compuesto suprime la duplicación de los retrovirus al inactivar la enzima transcriptasa inversa. Los análogos de los nucleósidos causan terminación prematura de la cadena de ADN provírico (precursor vírico). Todos estos inhibidores exigen fosforilación en las células anfitrionas antes de su incorporación al ADN vírico.
