Le réticulocyte se distingue du globule rouge mature par une membrane plus grande et légèrement festonnée et par des restes intracellulaires d'ARN qui subsistent après la perte du noyau. La coloration fait apparaître ces restes d'ARN sous forme de filaments formant un réseau irrégulier, auquel la cellule doit son nom.
