La première étape consiste à fabriquer un anticorps monoclonal capable de reconnaître la molécule étudiée (dans ce cas, le récepteur l'IL2). A cette fin, ces chercheurs immunisèrent des souris avec des lymphocytes T humains activés par un antigène, et recherchèrent les anticorps ne réagissant qu'avec les lymphocytes T activés et non avec les lymphocytes au repos. Ces anticorps furent baptisés "anti-TAC" (Anti T Activated Cells). Les expériences in vitro démontrèrent que les anticorps anti-TAC reconnaissent le récepteur de l'IL2, car ils inhibent la fixation de l'immuno-hormone et bloquent la prolifération des lymphocytes T activés.
