A somewhat different type of regulation of enzyme activity is the enzyme-catalyzed activation of inactive precursors of enzymes (zymogens) to yield the catalytically active forms. The classical examples are the digestive enzymes pepsin, trypsin, and chymotrypsin which are synthesized as the inactive zymogens pepsinogen, trypsinogen, and chymotrypsinogen, respectively. When these enzymes are secreted into the gastrointestinal tract, they are converted into their active forms by the selective hydrolytic cleavage of one or more specific peptide bonds in the zymogen molecule.... Trypsinogen is converted into active trypsin by removal of a hexapeptide from the N-terminal end, through the action of ... kinase.

In another application of gene therapy we have used an adenoviral vector that carries the gene for herpes simplex virus thymidine kinase (HSV-tk). Cells expressing HSV-tk will convert the anti-viral drug ganciclovir (GCV) to a form that is toxic to rapidly dividing cells. Using this adenoviral vector we have repeatedly shown robust tumoricidal effects in a variety of animal models of CNS tumor and will soon take this treatment to phase I clinical trials. We are extending this work to the manipulation of the CNSs immune response to tumors using adenoviral vectors that carry several cytokine genes.

Upon activation by binding double-stranded RNA in infected cells, the kinase down-regulates the cellular translational apparatus, and thus impairs viral protein expression. To overcome the inhibitory effects of PKR, viruses have developed efficient methods to prevent the activation or function of the kinase. A potential site of therapeutic intervention is to block viral inhibition of PKR.

Activation of macrophages by bacterial lipopolysaccharide (LPS) induces transcription of genes that encode for proinflammatory regulators of the immune response. Previous work has suggested that activation of the transcription factor activator protein 1 (AP-1) is one LPS-induced event that mediates this response. Consistent with this notion, we found that LPS stimulated AP-1- mediated transcription of a transfected reporter gene in the murine macrophage cell line RAW 264.7. As AP-1 activity is regulated in part by activation of the c-Jun N-terminal kinase (JNK), which phosphorylates and subsequently increases the transcriptional activity of c-Jun, we examined whether LPS treatment of macrophages resulted in activation of this kinase.

Insoluble peptidoglycan (PGN) from staphylococcal cell walls as well as soluble PGN (sPGN) secreted by staphylococci in the presence of beta-lactam antibiotics induced TNF- alpha mRNA and secretion of bioactive TNF-alpha in the murine RAW264.7 macrophage cell line. PGN and sPGN also induced rapid and dose-dependent tyrosine phosphorylation of several cellular proteins, including lyn and mitogen-activated protein kinases (extracellular signal-regulated kinases; but not hck, fgr, or vav) and increased the activities of mitogen-activated protein and rsk kinases. These PGN- and sPGN-induced effects were qualitatively similar to the effects induced by ReLPS, but higher concentrations of PGN and sPGN than ReLPS were required.
