Les acides nucléiques peptidiques (ANP) sont des homologues synthétiques des acides nucléiques, au niveau de leur composition. Cependant, les grandes différences entre ces deux derniers demeurent dans les propriétés physico-chimiques de ces molécules. La colonne dorsale des ANP n’est pas composée d’une queue de liaisons phosphodiesters, comme c’est le cas pour les acides nucléiques naturels. À la place, ils sont caractérisés par une queue protéique de liaisons peptidiques. En conséquence, la queue est d’une charge nette neutre et est dépourvue de carbones chiraux. Comme la queue polypeptidique d’un ANP n’est pas chargée de façon électronégative, il n’y a pas de répulsion électrostatique entre les deux brins d’un duplex. Nucleic acid testing, called NAT by blood banks, promises to close that gap. It could virtually eliminate the few cases of HIV that taint blood every year, said Dr. Celso Bianco of the New York Blood Center. En 1975 Sanger et Coulson conçoivent et élaborent une élégante méthode de détermination des séquences nucléotidiques de l'ADN, bien plus rapide que les méthodes utilisées précédemment pour le déchiffrement des séquences nucléotidiques de l'ARN. Le principe de la méthode, dite méthode plus et moins, est relativement simple. Il consiste à synthétiser un grand nombre de segments d'ADN, de longueurs différentes, et complémentaires à la séquence nucléotidique que l'on désire connaître.
