En RMN [résonance magnétique nucléaire] classique, tous les noyaux devraient résonner pour un couple déterminé (Bo, B1 : fréquence de radiation utilisée). Dans la réalité il faut tenir compte du phénomène de relaxation. En effet, après absorption de la radiation, le noyau a tendance à revenir à l'équilibre. On distingue deux sortes de relaxations : Relaxation spin-milieu ou spin-réseau, dite relaxation longitudinale. Ce phénomène longitudinal est provoqué par les molécules qui subissent un mouvement de rotation, de vibration, de translation et par les propriétés magnétiques des noyaux. L'énergie de translation est transférée au réseau du milieu. Relaxation spin-spin ou relaxation transversale.
