Lorsque Bruce Ames a ouvert la voie qui allait conduire aux tests actuels des cancérigènes potentiels, il a, parmi les diverses manifestations cellulaires consécutives aux lésions du DNA, choisi d'utiliser la mutagénèse des bactéries. Le tests d'Ames, aujourd'hui classique, combine l'usage d'une bactérie et celui d'un extrait de foie de rat. Prueba de Ames. Prueba de mutagenicidad en la cual una cepa de salmonella typhimurium mutante, caracterizada por su incapacidad para sintetizar histidina, se expone a un material que se va a ensayar, en un medio de cultivo que no contiene histidina. Si alguna de las bacterias recupera la capacidad de sintetizar la histidina y en consecuencia forma colonias, se comprueba que el material sometido a prueba tiene propiedades mutagénicas. The importance of the amino acid histidine lies in the fact that the body uses it to manufacture histamine, and histamine is responsible for a wide range of physiological processes. It is common knowledge that histamines cause the swelling and reddening in many inflammations and allergic reactions.
