The clonal selection theory is based on the proposition that during development each lymphocyte becomes committed to react with a particular antigen before ever being exposed to it. A cell expresses this commitment in the form of surface receptor proteins that specifically fit the antigen. The binding of antigen to the receptors activates the cell, causing it both to multiply and mature. Thus a foreign antigen selectively stimulates those cells that bear complementary antigen-specific receptors and are thus already committed to respond to it. This is what makes immune responses antigen-specific. The term "clonal" in clonal selection derives from the postulate that the immune system is composed of millions of different families, or clones, of cells, each consisting of T or B lymphocytes descended from a common ancestor. An encounter with a foreign pathogen or antigen stimulates naive T lymphocyte activation and the generation of immunologic memory. The T cells which are responsible for immunologic memory differ, both phenotypically and functionally, from naive T cells. Of interest is the observation that aging is associated with an enhanced level of memory T cells, suggesting that the qualitative T cell response differs in the elderly. Further, differences in the proportion of memory to naive T cells may be responsible for immune dysfunction and autoimmunity associated with the aging process. La fixation du V.I.H. à la cellule CD4+. La fixation de la particule virale sur la cible cellulaire se fait par interaction de la gp120 et de la molécule CD4, ce qui découvre la glycioritéine transmembranaire gp41. Une des extrémités de la gp41 s'introduit alors dans la membrane cellulaire et cet accrochage engendre la fusion entre la membrane virale et la membrane cellulaire. Le mécanisme de la pénétration intracellulaire du V.I.H. reste encore discuté. L'enveloppe virale conditionne donc l'infectivité du V.I.H. puisqu'il s'attache spécifiquement à la cellule qu'il infecte par l'intermédiaire de ses glycoprotéines d'enveloppe. Pour certains, la molécule CD4 n'interviendrait pas cependant comme un récepteur fonctionnel du V.I.H. Après la liaison spécifique de la gp120 à la molécule CD4 à la surface cellulaire, le virus pénétre dans la cellule.
