Any two chromosomes that undergo pairing during the prophase of meiosis. They are very similar in length and shape, have centromeres at the same point, and contain the same gene loci. They occur in all diploid organisms and their presence infers that two copies of each gene occur in each cell, although these copies may differ in being allelic forms of the same sequence.

Human artificial chromosomes (HAC) have been considered as a possible vector for human gene therapy. In our previous work a minichromosome (MC), isolated from a patient, was transformed with a selectable marker gene (neo) which was targeted into its centromere; the MC was then size reduced, by X-ray exposure, to be easily manipulated and further engineered. The method developed for gene targeting, based on the use of chromosome specific centromeric satellite DNA as transfection carrier, is highly efficient and reproducible.

In 1866, Down described clinical characteristics of the syndrome that now bears his name. In 1959, Lejeune and Jacobs et al. independently determined that Down syndrome is caused by trisomy 21, Down syndrome is by far the most common and best known chromosome disorder in humans. Mental retardation, dysmorphic facial features, and other distinctive phenotypic traits characterize the syndrome.

En 1866, le Docteur Langdon Down, fut le premier à décrire quelques-uns des traits des personnes atteintes [du syndrome qui porte son nom]. Il constata certains traits ressemblants au mongolien (habitant de Mongolie). C'est ainsi que le terme fut introduit dans notre vocabulaire. Puis en 1959, le Docteur Jérôme Lejeune découvrit que le «problème» se situait au niveau de la distribution des chromosomes. Il put vérifier que, sur la 21[e] paire, il y avait un chromosome de plus; c'est ce qui venait débalancer le système cellulaire. C'est pourquoi nous l'appelons aussi trisomie 21. Trisomie pour trois chromosomes et 21 pour la 21[e] paire.

OBS

Le syndrome de Down ou trisomie 21 est ce qu'on appelait autrefois «mongolisme». Aujourd'hui ce terme est rayé de notre vocabulaire étant donné sa connotation péjorative.

Les aberrations de structure sont la conséquence de cassures chromosomiques. Une cassure unique provoque la perte d'un fragment chromosomique : il y a délétion. Un cassure double sur un même chromosome peut entraîner la formation d'un chromosome en anneau qui résulte d'une cassure à chaque extrémité du chromosome suivie par la soudure des deux surfaces de section; ce chromosome en anneau équivaut à une double délétion. Si la cassure double intéresse deux chromosomes différents, les fragments rompus peuvent se recoller après échange réciproque et reconstituer des structures nouvelles : c'est une translocation.
