À chaque modification du champ, la molécule d'ADN doit se réorienter parallèlement au nouveau champ. Le temps nécessaire à la réorientation est proportionnel à la longueur de la molécule. Lorsque le champ est rétabli dans son sens initial, la molécule doit une nouvelle fois se réorienter. Ces temps de réorientation provoquent un retardement de la migration nette qui est proportionnel à la taille de la molécule. Le support de migration est un gel d'agarose à 1% et la taille des fragments séparés est de l'ordre de 50 kb à qq mégabases.

One current proteomic tool for visualizing and quantitating all proteins expressed in a ological system at a given time is two-dimensional gel electrophoresis (2-DGE). As originally described by Patrick O'Farrell for analyzing Escherichia coli proteins in 1975, 2-DGE combines the electrophoretic separation of denatured proteins bysoelectric point charge differences in the first dimension with separation based on molecular size differences in the second dimension. The proteins, which can be detected using protein-specific stains, appear as constellations of spots in the 2-D space of the gel. Over the 20-year history of 2-DGE, numerous algorithms have been developed for comparing 2-DGE patterns and quantitatively analyzing protein abundance.

Starch gel electrophoresis has been a popular technique, although it is now somewhat superseded by the use of polyacrylamide gels, which minimize convection and diffusion effects. A block or "plate" of starch gel is prepared, and the sample is applied in a narrow band (line) across the block about midway between the ends, which are contacted with electrodes through a connecting bridge. When current is passed through the cell, the different components of a mixture move with velocities that depend on their electric charges, their sizes, and their shapes. As electrophoresis proceeds, the negatively charged components migrate toward the anode and the positively charged components migrate toward the cathode. The result is a series of separated bands or lines of sample constituents, such as visualized by a stain.

Recently considerable progress has been made in assaying alleles, or variants, of some 15 genes that code for enzyme sysnthesis, which is readily detected by starch-gel electrophoresis.
