La limite théorique de résolution en microscopie optique classique (en champ lointain) suit le critère de Rayleigh, qui donne la résolution latérale optimale à 1.22 [lambda]/ (2n sin[(thêta)], soit environ 250 nm dans le spectre visible. Cette résolution peut être améliorée en augmentant l'ouverture angulaire et en utilisant un microscope à immersion pour augmenter l'indice de réfraction, et bien sûr en diminuant la longueur d'onde [lambda].
