Cette technique, initialement introduite par Castaing et Slodzian dès 1962 pour l'analyse de surface en géologie et en métallurgie, a été appliquée à la biologie par Galle à partir de 1970. Le principe de base de la méthode repose sur un principe simple; si une surface est bombardée par un faisceau d’ions primaires énergétiques ces particules vont induire la rupture des liaisons chimiques des molécules présentes dans les couches superficielles de l'échantillon et conduire à la désorption d’atomes ou de petits fragments polyatomiques neutres ou chargés(ions secondaires). Ces ions peuvent alors être accélérés et focalisés par des lentilles électrostatiques pour finalement être analysés et «triés» par un spectromètre de masse.
