Detergent solubilization has been used with great success to study the major proteins in various membranes. Typically, the membranes are first heated to 100°C in a 1% solution of the strong detergent SDS (which disrupts all non-covalent protein-protein and protein-lipid interactions and completely unfolds the proteins), then layered on top of a polyacrylamide gel containing SDS, and lastly subjected to electrophoresis in a strong electric field for several hours. They have no porosity except at a molecular level, but will absorb small amounts of certain liquids (such as solvents).
