It is a technique used in genetic studies that allows the detection and quantification of mRNA [messenger ribonucleic acid] It is a very sensitive method that shows whether or not a specific gene is being expressed in a given sample. RT-PCR is a very important test in the field of Genetically-Modified Organisms (GMO's) because it gives researchers a mechanism to test whether any specific gene is turned on (active) or turned off (inactive). This allows researchers to identify the benefits of genetically-modified organisms with respect to their "natural" counterparts and search for any significant differences in which genes are expressed in the two types of organisms. 3, record 10, English, - reverse%20transcription%2Dpolymerase%20chain%20reaction

A radically new approach—a molecular ticker tape—would, in one scenario, place a single strand of DNA with a known sequence of letters, or nucleotides, inside a cell but near its surface. An enzyme, DNA polymerase, would then add new nucleotides that bind to form a double-stranded molecule ... When a neuron fires, an influx of calcium ions coming through a newly opened membrane channel would cause the enzyme to add the wrong nucleotides ..., an error that could be detected when the DNA strand is later sequenced. 1, record 11, English, - molecular%20ticker%20tape

Until two years ago the nature of promoter base sequences was totally obscure. Now, however, a number of different promoter sequences are known and soon many more will be worked out. Soon after the initial binding to the RNA polymerase recognition site, the polymerase diffuses closer to the start of transcription to a group of At-rich base pairs to which it binds more tightly (RNA polymerase binding site). The fact that both binding sequences are AT-rich probably reflects the fact that such sequences denature much more easily than doGC-rich regions, and so are obvious targets for an enzyme like RNA polymerase, whose functioning demands local denaturation. 4, record 14, English, - AT%2Drich

Differential Display (DD) ... and a conceptually similar technique, arbitrarily primed PCR fingerprinting of RNA (APPCR)19, both PCR amplify subsets of an mRNA population and separate the resulting cDNAs by denaturing polyacrylamide gel electrophoresis. Since the introduction of DD in 1992, over 100 reports regarding improvements and/or successful applications have been published. Although DD has the appearance of being technically easy, the road from "band on the gel" to a positive can be treacherous as outlined in a recent commentary. The primary criticisms are (1) a high false positive rate, (2) questioned ability of DD to identify both abundant and rare mRNAs, (3) coding regions of mRNAs are usually not cloned, and (4) verification process is time consuming and usually requires a fair amount of RNA. 5, record 24, English, - differential%20display

The polymerase chain reaction (PCR) is a method of copying a DNA or RNA strand thousands of times, in a matter of minutes, by using the natural ability of an enzyme called polymerase to copy DNA. ... Scientists must know ... what DNA sequences flank or frame either ends of the desired portion. Complementary copies of the DNA sequences that flank the ends are created by using special enzymes; they are called primers. They identify the start and end of the copying process. ... Subchromosal genome rearrangements in cancer and alu-family DNA repeats. Evidence are accumulating suggesting existence in cancer cells of subchromosomal genome rearrangements, deletions and amplifications, covering up to several hundreds kb which are not followed by visible chromosome aberrations. Such interstitial micro-deletions and amplifications (IMDA) were found in leukemias and in solid tumors of different origin and they possibly manifest early changes in neoplasias which may be useful in diagnostics. In acute leukemias we have detected variations in localization of Alu clusters in chromosomes 1, 3, 6, 8 and 14 probably associated with IMDA but the role of Alu repeats in the promotion of these variations remains unclear. 1, record 39, English, - ALU%20family

... pN acts as an antitermination factor at transcription terminators. Evidence now exists for the presence of antitermination sequences within the E. coli chromosome; for example, just past the promoter for rrnG which encodes a rRNA. Nothing is known about how antitermination works in E. coli at this point. However, this suggests that even in the absence of bacteriophage lambda, antitermination is believed to control the level of the transcripts for genes critical to cell growth. This suggests a way that lambda can commandeer the cell by using its pN to increase the levels of primase and sigma 70 for its own replication and gene expression. 2, record 47, English, - antitermination%20factor

Primed in situ labeling (PRINS) is a fast alternative to traditional in situ hybridization, requiring only a tew handling steps (1,2). In the PRINS reaction, an unlabeled sequence-specific oligonucleotide primer or short DNA fragment is hybridized (annealed) to denatures chromosomal DNA and extended in situ in the presence of hapten-labeled nucleotide triphos phates by a suitable DNA polymerase (3). The technique is especially suited for the rapid identification of chromo somes with oligonucleotide primers specific for individual chromosomal sequences and chromosomal elements like centromer or satellite sequences. 2, record 49, English, - primed%20in%20situ%20labeling%20of%20nucleic%20acids

Short stretches of RNA have been found attached to the 5' ends of many newly synthesized DNA chains. The starting points for DNA replication may therefore be recognized not by a DNA polymerase, but by a form of RNA polymerase. Following binding of a starter RNA polymerase, a number of ribonucleotides are sequentially linked together until a stop signal is read. The enzyme then detaches leaving a short RNA chain still bound to its DNA template. This RNA chain then serves as a primer onto which DNA polymerase III adds deoxynucleotides. Removal of the RNA primer occurs later through the action of other enzyme(s) which can digest away the RNA component of RNA/DNA hybrids. One enzyme that has this capacity is DNA polymerase I, whose primary task may be to close the gaps between RNA-primed DNA fragments. 1, record 66, English, - RNA%2Dprimed%20DNA%20fragment

Experiments have demonstrated that the level of gene expression is influenced principally by the number of gene copies and the efficiency of their transcription and translation.
