Detergent solubilization has been used with great success to study the major proteins in various membranes. Typically, the membranes are first heated to 100°C in a 1% solution of the strong detergent SDS (which disrupts all non-covalent protein-protein and protein-lipid interactions and completely unfolds the proteins), then layered on top of a polyacrylamide gel containing SDS, and lastly subjected to electrophoresis in a strong electric field for several hours. Friction stir welding uses a cylindrical, shouldered tool with a profiled pin that is rotated and slowly plunged into the joint line between two pieces of sheet or plate material, which are butted together. Frictional heat is generated between the wear resistant welding tool and the work piece. This heat causes the work piece material to soften without reaching the melting point and allows the tool to traverse the weld line. As it does, the plasticized material is transferred from the leading edge of the tool to the trailing edge of the tool shoulder and pin. Le soudage par friction-malaxage(SFM) utilise le même principe [que le soudage par friction] pour joindre des plaques bout à bout ou avec chevauchement. L'outil utilisé est de forme cylindrique(profiled pin) avec un épaulement(tool shoulder). Le tout est mis en rotation à une vitesse de quelques centaines de tours par minute. Alors que l'outil approche les plaques, la rotation génère assez de chaleur pour ramollir le métal, sans jamais le rendre liquide, afin de permettre à l'outil de pénétrer dans les plaques. Une fois l'épaulement bien appuyé, assez de chaleur est générée pour permettre à l'outil de suivre une trajectoire de translation en suivant le joint entre les deux plaques et ainsi les souder. Les opinions concernant le terme «pressurisation» sont assez partagées. En effet, bien que ce terme soit largement utilisé et solidement ancré dans l'usage, le Comité d’étude des termes techniques français recommande d’en proscrire l'emploi. Ce Comité préconise plutôt le terme «mise en pression». Or, voici que la question se complique. En effet, des spécialistes en aéronautique(CUTA maintenance) établissent une distinction de sens entre ces deux termes :«pressurisation» étant réservé pour la cabine et «mise en pression» pour les circuits. Quoi qu'il en soit c'est à l'usage qu'il faut s’en remettre et pour l'instant, cette distinction ne semble pas faire l'objet d’un consensus, pas plus d’ailleurs que la recommandation du Comité. Student Challenge: Clamp one end of a 30 cm steel ruler to the edge of a table so that it sticks straight out. Bend the free end of the ruler downward a few centimetres and release it. Observe what happens. Attach a heavy mass (approximately 200 grams), such as a large blob of putty or Plasticine, to the free end of the ruler.
