La première méthode utilisée pour repérer la position réelle de différentes mutations du bactériophage lambda est basée sur la possibilité de fragmenter mécaniquement l'ADN en des segments de taille définie. Les fragments qui ont une densité différente peuvent être séparés les uns des autres par centrifugation à l'équilibre dans un gradient de chlorure de césium. Les fragments riches en GC ayant une densité plus grande que les fragments riches en AT, la séparation devient une opération de routine si les différents segments possèdent des rapports AT/GC distincts. Some viruses may remain dormant inside host cells for long periods, causing no obvious change in their host cells (a stage known as the lysogenic phase). Certains bactériophages(...) ont la propriété lorsqu'ils infectent une bactérie d’intégrer un gène bactérien dans leur génome(...). Un exemple classique est le bactériophage lambda, qui, au cours de sa multiplication dans le colibacille, intègre parfois dans son génome un gène du colibacille, le gène galactose par exemple. Lorsque le phage lambda se multiplie dans la bactérie, il se forme cent à mille copies du génome du phage dans chaque bactérie parasitée. Le gène galactose porté par le phage est alors amplifié d’autant. On peut considérer, et ceci est une notion nouvelle par rapport au clonage cellulaire, que grâce au phage il y a eu clonage du gène galactose. En effet, grâce au phage transducteur il y a eu isolement d’un gène(le gène galactose), puis amplification de ce gène. Toute la population(ou clone) de phage transducteur contient ce même gène galactose.
